Due to extensive and repeated use of insecticide, this species has developed many different resistance mechanisms to insecticides. The enzyme acetylcholinesterase (EC 3.1.1.7) (AChE) occurs widely in both vertebrate and invertebrate nervous systems and is the target of organophosphate (OP) and carbamate insecticides. One of resistance mechanisms to OP and carbamate insecticides is the insensitivity of AChE to these insecticides. In this study the possibility of mutation in insensitive AChE was surveyed. The complete coding sequences of acetylcholinesterases (MpAChE2) from susceptible and resistant populations were identified and sequences from resistant and two susceptible populations were compared. Consequently, one amino acid substitution (Ser431Phe) was detected within the resistant population at MpAChE2 gene. The Ser431Phe substitution is located in acyl pocket of acetylcholinesterase. Therefore, it seems that MpAChE2 is important as a target of pirimicarb. This substitution makes the acyl pocket narrower and pirimicarb can not have any access to this site. cDNA was obtained from individual aphids randomly and sequence of MpAChE2 indicated that MpAChE2 of the resistant population was heterozygous.}, keywords = {Myzus persicae,resistance to insecticide,insensitive acetylcholinesterase,point mutation}, title_fa = {مقاومت به حشره‌کش‌های فسفره و پیریمیکارب ناشی از جابجایی اسیدآمینه‌ی Ser431Phe در آنزیم استیل‌کولین‌استراز شته‌ی سبز هلو، Myzus persicae (Hem.: Aphididae)}, abstract_fa = {شته‌ی سبز هلو، Myzus pesicae Sulzer یکی از آفات مهم گیاهان زراعی و زینتی در سرتاسر جهان می‌باشد. به دلیل استفاده‌ی مکرر و گسترده از ترکیبات شیمیایی، این آفت ساز و کارهای مختلف مقاومت به حشره‌کش‌ها را بروز داده است. آنزیم استیل‌کولین‌استراز (EC 3.1.1.7) (AChE) به‌طور وسیع در سیستم عصبی مهره‌داران و بی‌مهره‌گان وجود دارد و محل هدف دو گروه اصلی از حشره‌کش‌ها یعنی ترکیبات آلی فسفره و کاربامات‌هاست. یکی از مکانیسم‌های درگیر در مقاومت به آفت‌کش‌های فسفره و کاربامات، آنزیم استیل‌کولین‌استراز غیرحساس به این ترکیبات است. در این تحقیق احتمال جهش در آنزیم استیل‌کولین‌استراز غیرحساس به پیریمیکارب مورد بررسی قرار گرفت. توالی‌های رمزگردان کامل ژن استیل‌کولین‌استراز MpAChE2 جمعیت مقاوم و دو جمعیت حساس به پیریمیکارب شناسایی و با هم مقایسه شدند. نتایج نشان داد که در MpAChE2 اسیدآمینه‌ی فنیل‌آلانین جایگزین اسیدآمینه‌ی سرین 431 شده و این جابجایی در بخش اسیل آنزیم واقع شده است. بنابراین به نظر می‌رسد که MpAChE2 نقطه‌ی هدف حشره‌کش پیریمیکارب باشد و این جابجایی اسید‌آمینه باعث شده اسیل‌ پاکت باریک‌تر شده و پیریمیکارب نتواند در نقطه‌ی هدف جای گیرد. برای بررسی دقیق هتروزیگوت یا هموزیگوت بودن جهش ایجاد‌شده، cDNA از حشرات انفرادی به طور تصادفی تهیه گردید. نتایج نشان داد که MpAChE2 حشرات انفرادی همگی در مکان جهش هتروزیگوت بودند.}, keywords_fa = {Myzus persicae,مقاومت به حشره‌کش‌ها,استیل‌کولین‌استراز غیرحساس,جهش نقطه‌ای}, url = {https://jesi.areeo.ac.ir/article_105237.html}, eprint = {https://jesi.areeo.ac.ir/article_105237_29a0d760ccd04ef692e532097b0774fb.pdf} }
